VIASURE EBV q Real Time PCR Detection Reagents RUO is a qPCR test designed for the quantitative detection of EBV DNA in human EDTA-plasma samples. After DNA isolation, the detection of EBV is performed by the amplification of a conserved region of the BNRF1 gen, using specific primers and fluorescent-labelled probes.
VIASURE EBV q Real Time PCR Detection Reagents RUO is based on the 5´exonuclease activity of DNA polymerase. During DNA amplification, this enzyme cleaves the probe bounded to the complementary DNA sequence, separating the quencher dye from the reporter. This reaction generates an increase in the fluorescent signal which is proportional to the quantity of target template. This fluorescence can be measured on Real Time PCR platforms.
VIASURE EBV q Real Time PCR Detection Reagents RUO contains in each well all the components necessary for real time PCR assay (specific primers/probes, dNTPs, buffer and polymerase) in a stabilized format, as well as an endogenous internal control to monitor the extraction process and/or discard the inhibition of the polymerase activity. The assay uses a human housekeeping gene as an endogenous internal control (EIC) (human RNase P gene). Human housekeeping genes are involved in basic cell maintenance and, therefore, are expected to be present in all nucleated human cells and maintain relatively constant expression levels.
In addition, this kit contains 5 lyophilized vials (EBV1Q1 to EBV1Q5) with different standardized concentrations of EBV specific DNA (EBV q Quantitative Standards) for the quantification of EBV viral DNA in research samples. These quantitative standards are calibrated against the 1st WHO International Standard for EBV DNA for Nucleic Acid Amplification Techniques (NIBSC code 09/260), ensuring the quality of the results and the comparison with results from other laboratories.
